To investigate the relationship between AQP11 expression and oocyte quality, ten AQP subtypes were quantified in the ovarian cortex and follicles from PCOS patients and non-PCOS patients using RT-PCR (Figure 8). Interestingly, the mRNA levels of AQP11 were significantly lower in oocytes of patients with PCOS than in those of non-PCOS oocytes. Furthermore, AQP11 mRNA was positively correlated with LHR and negatively correlated with StAR, indicating that the protein is involved in the steroidogenesis of the oocytes.
We next investigated the expression of AQP11 in the mouse brain at different postnatal days to observe the developmental change. AQP11 was expressed at the surface of the pia matter but not at the capillaries in early postnatal stages from P1 to P14. At the stage of P28, AQP11 was mainly expressed at the capillaries and less at the pia matter (Figure 3A,B).
In addition, AQP11 staining partially overlapped with the staining for glucose transporter 1 (GLUT1) in the brain capillary endothelium (Figure 2F), suggesting that AQP11 is also involved in BBB water transport. In AQP11-deficient mice, ER-CAT expression was reduced (Figure 4A,B), but the levels of CTF, which is required for a functional PC-1 molecule, were largely unchanged, suggesting that AQP11 does not affect auto-processing of the precursor to PC-1.
